Q: How small an oligonucleotide can be recovered? A: The Agarose Gel kit can handle DNA as short as 20 nt. Recovery percentages get better with larger sizes. 50 to 200 bp gives recovery of 70% or better. 200+ bp gives recovery of 90% or better. The Poly-Gel kits have similar capabilities. For dsDNA, fragments won't anneal well if they're shorter than about 20 nt anyway. For ssDNA, you should be able to recover fragments less than 20 nt, perhaps down to as few as 7 or 8. This will require a high gel concentration (i.e., firmness). |

#DC3511. Agarose Gel Extraction Kits: DNA fragments from 20 bp up to 70 kb can be purified with the kit. Can purify DNA fragments from RFLP, PCR, phosphorylation, labeling, hybridization, ligation, and other enzymatic reactions. The EZBindtm mini-column can give over 80-90% recovery.

#DC3512. Polyacrylamide Gel Extraction Kits (for DNA): recover ssDNA (as short as 7 or 8 bases) or dsDNA after PAGE. Visualize the oligonucleotides in the gel using ethidium bromide or UV shadowing, then cut out the bands and soak. The DNA is eluted using gel elution buffer. Special binding buffer is then added to facilitate binding of the DNA fragments to silica matrix. Impurities (e.g., protein, salts) are removed with wash buffer; DNA is eluted from matrix using sterile deonized H2O or low salt buffer.
Recovered DNA is suitable PCR, hybridization, ligation, and other procedures. Spin column format allows parallel processing of multiple samples
Extraction kit works with urea-denaturing PAGE.
